The maintenance of metabolic health requires complex regulation of metabolic processes in metabolic tissues. The coordination of this metabolic regulation involves extensive crosstalk among tissues. Signaling factors that are secreted into the circulation and impart systemic metabolic effects include molecules such as the liver secreted hepatokines and lipid mediators. Many of these factors regulate lipid metabolism, including de novo lipogenesis. For instance, monounsaturated fatty acids, such as palmitoleate and oleate, are major products of de novo lipogenesis and exert significant control over metabolic regulation. These fatty acids may circulate as free or esterified forms and subsequently influence metabolic processes in tissues distinct from the one in which they are synthesized. Examples include the effects of hepatic palmitoleate and oleate in adipose tissue and of adipose tissue-derived palmitoleate in liver. The updated edition utilizes the vast literature and ongoing research on liver function to focus on liver de novo lipogenesis and its role in systemic metabolic processes and signaling. Each chapter has been thoroughly updated for this new edition.

James M. Ntambi, PhD, is the professor of Biochemistry and the Steenbock Professor of Nutritional Sciences in the College of Agricultural and Life Sciences, at the University of Wisconsin, Madison. He is also an Adjunct Professor of Physiology, Pharmacology and Therapeutics, Johns Hopkins University School of Medicine, Baltimore, MD, USA. Professor Ntambi has more than four decades of teaching, mentoring, and research experience in biochemistry, molecular biology, nutrition, and metabolism. His research focuses on the genetic regulation of metabolism, adipocyte biology and differentiation. He has worked on hormonal and dietary regulation of gene expression, and in more recent years has used a multidisciplinary approach to unravel the physiological role of the stearoyl-CoA desaturase genes in lipid and carbohydrate metabolism. He is studying the influence of dietary and endogenously synthesized monounsaturated fatty acids in de novo lipogenesis, obesity, diabetes, insulin resistance, inflammation, fatty liver disease and cancer. He has authored more than 250 scientific publications and numerous book chapters and reviews. His honors include the 2013 ASBMB) Award for Exemplary Contributions to Education, 2023 Fellow of ASBMB and the James M. Ntambi Postdoctoral Fellowship in Biomedical Discovery established by the department of Biochemistry at the University of Wisconsin-Madison in his honor.
