Role of Lysosomal Functions in Health and Disease

Fulvio Reggiori

Science / Life Sciences / Biochemistry

This comprehensive book explores the multifaceted world of lysosomes, describing their critical roles in cellular functions and homeostasis, and human health. It begins with the fascinating journey of lysosome discovery, tracing early observations to groundbreaking research that unveiled their biogenesis and maintenance as well as their importance in cellular processes.

Readers will learn about the intricate pathways involved in lysosome formation and the transport mechanisms that ensure their proper function, including endocytosis and autophagy. The dynamic nature of lysosomes is highlighted, focusing on their ability to fuse with other organelles and reform, which is essential for maintaining their own homeostasis and ultimately that of cells.

The book explores how lysosomes play a pivotal role in sensing nutrients and regulating cellular metabolism, ensuring the balance and health of the cell. It delves into the genetic and transcriptional networks that govern lysosome maintenance and functionality, emphasizing the complexity of their regulation. Understanding the significance of lysosome positioning within cells and how their localization impacts their functional roles is another key aspect. Specialized regions where lysosome membranes interact with other organelles are discussed, facilitating communication and coordination within the cell.

The variation of lysosomal functions across different cell types is examined, highlighting the diversity and specialization of these organelles. The role of lysosomal ion channels in regulating cellular physiology, including their impact on ion homeostasis and signal transduction, is explored. Mechanisms by which cells respond to lysosomal damage, including repair processes and lysophagy, a specialized form of autophagy, are discussed. The consequences of lysosomal dysfunction are linked to various diseases such as neurodegenerative disorders and lysosomal storage diseases, as well as the aging process. Finally, the potential of targeting lysosomes for therapeutic interventions is explored, offering hope for treating diseases linked to lysosomal dysfunction.

This book is an essential resource for researchers, students, and professionals interested in cellular biology, pathology, and therapeutic development. It provides a thorough understanding of lysosomal functions beyond the degradation of intracellular and extracellular material, and their implications for health and disease.
